目的 :观察前列腺素 E2 (PGE2 )对去卵巢 2周大鼠骨骼的影响。方法 :3月龄 Sprague- Dawley大鼠 2 4只随机分为 3组 :(1 )假手术组 ;(2 )去卵巢组 ;(3)前列腺素 E2 (预防组 )。预防组用 PGE2 0 .6 mg· kg- 1 · d- 1 皮下注射。全部大鼠于实验 2周时处死 ,取胫骨上段不脱钙骨制片后测量。结果 :(1 )与假手术组比较 ,去卵巢大鼠 2周已出现有统计学意义的骨丢失 ,减少 46% ,伴有骨高转化率的改变 ,即代表骨转化率的参数 (Act.F)增加。 (2 )与去卵巢模型组比较 ,PGE2 组骨量增加 ,骨形成 ,骨吸收的参数和代表骨转化率的参数 (Act.F)维持在去卵巢的高水平。结论 :合成代谢药 PGE2 能预防去卵巢 2周大鼠的骨丢失 ,表现骨高转化的改变。
Objective: To observe the effect of prostaglandin E2 (PGE2) on the skeletal structure of 2-week ovariectomized rats. Methods: Twenty four 3-month-old Sprague-Dawley rats were randomly divided into three groups: (1) sham operation group, (2) ovariectomized group, and (3) prostaglandin E2 (prophylaxis group). Prevention group with PGE2 0. 6 mg · kg-1 · d-1 subcutaneous injection. All rats were sacrificed 2 weeks after the experiment, taking the upper tibia non-decalcified bone tablets measured. Results: (1) Compared with the sham operation group, there was a statistically significant loss of bone loss in ovariectomized rats for 2 weeks, with a 46% reduction, accompanied by a change in bone turnover rate, which represents the parameter of bone turnover rate (Act. F) increase. (2) Compared with the ovariectomized model group, the bone mass, bone formation, bone resorption parameters and the parameters representing the bone turnover rate (Act.F) in PGE2 group were maintained at the high level of ovary. Conclusion: The anabolic agent PGE2 can prevent the bone loss in ovariectomized rats for 2 weeks and show the change of bone hyperplasia.
