目的:探讨hTERTmRNA及hTERT蛋白在口腔颌面部肉瘤组织中表达的生物学意义。方法:应用原位杂交和免疫组化技术检测37例口腔颌面部肉瘤和14例良性间叶组织肿瘤中hTERTmRNA和hTERT蛋白的表达,探讨口腔肉瘤组织中hTERT表达的生物学意义。结果:肉瘤中hTERTmRNA的阳性表达率明显高于良性肿瘤(62.2%/14.3%,P<0.05)。在低度、中度和高度恶性肉瘤组织中,hTERTmRNA阳性表达率分别为41.67%、50.00%和86.67%,统计分析显示低度恶性与高度恶性组间差异有统计学意义(P<0.05)。复发肉瘤中hTERTmRNA的阳性表达率明显高于原发肉瘤(P<0.05)。在发生淋巴结转移或远处转移的肉瘤中hTERTmRNA表达有升高趋势(P>0.05);口腔肉瘤中hTERT蛋白的阳性表达率明显高于良性肿瘤(P<0.05),并随肉瘤分化程度的降低而呈升高趋势,其中低度恶性肉瘤与高度恶性肉瘤比较差异显著(P<0.05)。hTERT蛋白表达与肉瘤复发密切相关(P<0.05),发生淋巴结转移或远处转移的肉瘤中hTERT蛋白表达明显升高(P>0.05)。结论:hTERT基因在口腔颌面部肉瘤发生与发展过程中可能起重要作用。
Objective: To investigate the biological significance of hTERT mRNA and hTERT protein expression in oral and maxillofacial sarcoma. Methods: The expressions of hTERT mRNA and hTERT protein in 37 cases of oral and maxillofacial sarcoma and 14 cases of benign mesenchymal tumor were detected by in situ hybridization and immunohistochemistry, and the biological significance of hTERT expression in oral sarcoma was explored. Results: The positive rate of hTERT mRNA in sarcoma was significantly higher than that in benign tumor (62.2% / 14.3%, P <0.05). The positive rates of hTERT mRNA in low, moderate and high grade sarcomas were 41.67%, 50.00% and 86.67%, respectively. Statistical analysis showed that there was significant difference between low grade and high grade malignant tumors (P <0.05). The positive rate of hTERT mRNA expression in recurrent sarcoma was significantly higher than that in primary sarcoma (P <0.05). The expression of hTERT mRNA in sarcomas with lymph node metastasis or distant metastasis had an increasing trend (P> 0.05). The positive rate of hTERT protein in oral sarcoma was significantly higher than that in benign tumors (P <0.05), and decreased with the differentiation of sarcoma But showed a trend of increase. There was significant difference between low grade sarcoma and high grade sarcoma (P <0.05). The expression of hTERT protein was closely related to the recurrence of sarcoma (P <0.05). The expression of hTERT protein in sarcoma with lymph node metastasis or distant metastasis was significantly increased (P> 0.05). Conclusion: hTERT gene may play an important role in the occurrence and development of oral and maxillofacial sarcoma.
