目的制备脑组织特异性表达胰岛素样生长因子-1(Insulin-Like Growth Factors 1,IGF-1)转基因小鼠。方法采用精子为载体法进行基因转导,出生后小鼠PCR检测,建立转基因小鼠系,用Western blot和免疫荧光法分别检测转基因小鼠海马齿状回亚粒状区(subgranular zone,SGZ)IGF-1表达量和报告基因EGFP阳性细胞数。结果获得3只阳性小鼠,2只外源基因能稳定遗传,并建立了转基因小鼠系,转基因鼠SGZ区域IGF-1表达量显著高于正常鼠,EGFP也在此区域表达。结论成功制备IGF-1转基因小鼠。
Objective To prepare brain tissue to express insulin-like growth factor-1 (IGF-1) transgenic mice. Methods Spermatozoa were used as vectors for gene transduction. After birth, mice were detected by PCR and transgenic mice lines were established. Western blot and immunofluorescence were used to detect the expression of subgranular zone (SGZ) IGFs in hippocampus of transgenic mice -1 expression and reporter EGFP positive cells. Results Three positive mice were obtained, and two foreign genes were stably inherited. A transgenic mouse line was established. The expression of IGF-1 in SGZ of transgenic mice was significantly higher than that in normal mice, and EGFP was also expressed in this region. Conclusion IGF-1 transgenic mice were successfully prepared.
