目的:建立脑卒中后抑郁症(post-stroke depression,PSD)的大鼠模型,观察海马脑源性神经营养因子(BDNF)的损伤情况,并从分子生物学角度研究中药涤痰开窍方对脑卒中后抑郁症大鼠海马区BDNF蛋白表达的影响及神经保护作用。方法:采用双侧颈总动脉永久性结扎致大鼠慢性低灌注PSD模型,研究涤痰开窍方对脑卒中后抑郁症大鼠的治疗作用,采用用免疫组化法检测大鼠海马BDNF蛋白的表达。结果:与模型组比较,涤痰开窍方能使海马C3区BDNF蛋白表达增多(P<0.01或<0.05)。结论:中药涤痰开窍方的治疗能上调卒中后抑郁症大鼠海马BDNF蛋白,从而起到保护神经元的作用。
OBJECTIVE: To establish a rat model of post-stroke depression (PSD) and to observe the damage of brain-derived neurotrophic factor (BDNF) in hippocampus. To study the effect of Ditanqiqiao decoction on brain Effect of BDNF protein expression in the hippocampus of rats with post - stroke depression and its neuroprotective effects. Methods: Chronic low perfusion PSD rat model of bilateral common carotid artery ligation was used to study the therapeutic effect of Ditanqikaiqiao decoction on post-stroke depression in rats. Immunohistochemistry was used to detect the expression of BDNF protein expression. Results: Compared with the model group, the expression of BDNF protein in the C3 area of hippocampus was increased with the Phlegm Resuscitation (P <0.01 or <0.05). Conclusion: The treatment of Ditanqikaiqiao decoction can up-regulate the BDNF protein in the hippocampus of post-stroke depression rats, thus protecting the neurons.
