目的采用UPLC法测定半夏药材及其炮制品中草酸钙的含量。方法色谱柱为Acquity UPLC BEH C_(18)柱(50 mm×2.1 mm,1.7μm),流动相为0.5%(NH_4)H_2PO_4缓冲液(磷酸调pH2.8),流速0.5 m L·min~(-1),柱温30℃,检测波长210 nm。结果线性方程为:Y=1.8488×10~4X+4.146×10~3(r=0.9999),2.58~25.80μg·m L~(-1)草酸与峰面积的线性关系良好,平均加样回收率为98.58%,RSD=1.01%(n=9)。结论所用方法准确可靠,可为半夏药材的质量控制提供参考。
Objective To determine the content of calcium oxalate in Pinellia ternata and its processed products by UPLC method. Methods The mobile phase was Acity UPLC BEH C 18 column (50 mm × 2.1 mm, 1.7 μm). The mobile phase consisted of 0.5% (NH 4) H 2 PO 4 buffer (adjusted to pH 2.8 with phosphoric acid) and the flow rate was 0.5 m L · min ~ -1), column temperature 30 ℃, detection wavelength 210 nm. The linear equation was: Y = 1.8488 × 10 ~ 4X + 4.146 × 10 ~ 3 (r = 0.9999). The linear relationship between peak area and concentration of oxalic acid was 2.58 ~ 25.80μg · m L -1. 98.58%, RSD = 1.01% (n = 9). Conclusion The method is accurate and reliable, which can provide reference for the quality control of Pinellia ternata.
