目的对在临床治疗上有较好疗效的柴芪汤进行实验研究,探讨其干预代谢综合征(MS)的作用机制。方法采用高脂高糖高盐饮食喂养方法制备大鼠代谢综合征模型,并分为模型组、中药组、罗格列酮组和正常组。中药组从造模开始即给予柴芪汤,给药量为5.71 g/(kg.d);罗格列酮组从造模开始给予罗格列酮1.2 mg/(kg.d)。给药剂量按大鼠的体表面积进行计算。观察并比较不同组别大鼠的一般情况、实验室指标及肝脏组织学变化。用生化法检测丙氨酸氨基转移酶(ALT)、甘油三脂(TG)、空腹血糖(FBG),放免法检测空腹胰岛素(FINS)、硫代巴比妥酸法测定肝匀浆苯二醛(MDA)、黄嘌呤氧化酶法测定肝匀浆超氧化物歧化酶(SOD)、HE染色法观察肝脏的病理组织学改变。结果模型组大鼠一般情况逐渐变差,第8周代谢综合征造模成功。中药组及罗格列酮组肝功能、血脂、血糖、胰岛素敏感性、肝组织病理形态学改变及SOD活性、MDA含量等指标均优于模型组。中药组在改善TG、降糖及改善胰岛素敏感性方面稍逊于罗格列酮组,余指标改善情况均优之,但2组比较无统计学差异。结论柴芪汤对代谢综合征和非酒精性脂肪肝(NAFLD)均有较好的防治作用,既可以降低血清中胰岛素含量,改善胰岛素抵抗(IR)的状态,又可以改善脂代谢,可能是治疗MS和NAFLD的作用机制之一。
Objective To study the effect of Chaiqi Decoction with good curative effect on clinical treatment and explore the mechanism of intervention on metabolic syndrome (MS). Methods Metabolic syndrome model was induced by high-fat, high-sugar and high-salt diet. The rats were divided into model group, TCM group, rosiglitazone group and normal group. The traditional Chinese medicine group was given Qianqi Decoction at the dosage of 5.71 g / (kg · d). Rosiglitazone group was given Rosiglitazone 1.2 mg / (kg · d). The dose is calculated on the body surface of the rat. Observed and compared the general situation of different groups of rats, laboratory and liver histological changes. The levels of ALT, TG, FBG and FINS in serum were determined by biochemical methods. The plasma phthalaldehyde was detected by thiobarbituric acid method. (MDA), and superoxide dismutase (SOD) in liver homogenate were detected by xanthine oxidase method. The pathological changes of liver were observed by HE staining. Results The general condition of rats in the model group gradually became worse, and the metabolic syndrome was successfully established in the 8th week. The liver function, blood lipid, blood glucose and insulin sensitivity of the TCM group and rosiglitazone group were better than those of the model group in pathological morphological changes, SOD activity and MDA content. Chinese medicine group in the improvement of TG, hypoglycemic and improve insulin sensitivity less than rosiglitazone group, the improvement of the remaining indicators are excellent, but there was no significant difference between the two groups. Conclusions Chaiqi Decoction has good preventive and therapeutic effects on metabolic syndrome and nonalcoholic fatty liver disease (NAFLD).
