雌激素受体基因(ESR)、促卵泡素β亚基基因(FSHβ)和瘦素基因(leptin)是影响卵泡生长发育的主效基因,在母猪繁殖过程中起关键的作用。试验采用PCR和PCR-RFLP技术检测ESR、FSHβ和leptin基因的单核苷酸多态性,分析各基因多态性与卵泡囊肿的关系。结果表明:3个基因共检测到3个多态位点,分别是位于ESR基因内含子A>G突变,FSHβ基因内含子T>C突变,leptin基因外显子C>T突变。ESR的AA和AB基因型在健康群体和卵泡囊肿群体分布差异显著(P<0.05),该基因型可以作为检测卵泡囊肿的标志基因型;FSHβ基因和leptin基因的AA、AB和BB基因型分布在检测群体中均未见显著差异(P>0.05)。
Estrogen receptor gene (ESR), follicle stimulating hormone (FSHβ) gene and leptin (leptin) are the main effect genes that affect the growth and development of follicles and play a key role in sow breeding. The single nucleotide polymorphisms of ESR, FSHβ and leptin gene were detected by PCR and PCR-RFLP. The relationship between polymorphisms and follicular cysts was analyzed. The results showed that three polymorphic loci were detected in the three genes, which were intron A> G mutation of ESR gene, intron T> C mutation of FSHβ gene and exon C> T mutation of leptin gene respectively. The distribution of AA and AB genotypes in ESR was significantly different between healthy group and follicular cyst population (P <0.05), which could be used as a marker genotype for detecting follicular cyst.
