目的研究多种胃癌易感性相关细胞因子基因单核苷酸多态性(single nucleotide polymorphisms,SNPs)在慢性萎缩性胃炎(chronic atrophy gastritis,CAG)患者中的分布特征及两者的相关性。方法根据文献最新报道,选择胃癌易感性相关基因SNP位点:IL1-1B-311/-511、IFN-γ+874、IL-8-251、IL-10-1082/-819/-592、TNF-α-308/-857、MIF-173等6个基因上10个SNP位点。采用PCR扩增基因组DNA,直接测序法对30例CAG及30例健康对照的各SNP进行分析。结果IL-10-592位点三种基因型在CAG和健康对照组间的分布差异有统计学意义,χ2=6.83,P=0.03。其余9个SNP基因型、所有等位基因型在CAG和健康对照组间的分布差别均无统计学意义。结论CAG患者中携有多个胃癌易感性相关细胞因子基因SNP,但除IL-10-592CC基因型可能与CAG相关,其余SNP与CAG未见相关性。
Objective To investigate the distribution of single nucleotide polymorphisms (SNPs) of several gastric cancer susceptibility related cytokines in patients with chronic atrophy gastritis (CAG) and their relationship. Methods According to the latest reports in the literature, SNPs of gastric cancer susceptibility genes were selected: IL1-1B-311 / -511, IFN-γ + 874, IL-8-251, IL- 10-1082 / -819 / -592, TNF -α-308 / -857, MIF-173 and other 6 genes on the 10 SNP sites. Genomic DNA was amplified by PCR, and the SNPs of 30 CAGs and 30 healthy controls were analyzed by direct sequencing. Results There was significant difference in the distribution of IL-10-592 genotypes between CAG and healthy controls (χ2 = 6.83, P = 0.03). The other nine SNP genotypes, the distribution of all the alleles in CAG and healthy controls were not statistically different. Conclusions There are multiple SNPs associated with gastric cancer susceptibility in CAG patients. However, the genotypes of IL-10-592CC may be related to CAG, and the rest of SNPs have no correlation with CAG.
