目的观察大蒜素对血管紧张素II诱导血管平滑肌细胞增殖的抑制作用。方法采用培养的兔血管平滑肌细胞,应用3H-TdR掺入法,观察在血管紧张素II促进VSMC增殖过程中,大蒜素对血管平滑肌细胞DNA合成的影响及其时间效应。结果血管紧张素II可促进处于静止状态的兔血管平滑肌细胞DNA的合成,36h时细胞DNA的合成达到高峰。大蒜素对血管紧张素II诱导的血管平滑肌细胞增殖有明显的抑制作用,并呈现出明显的浓度依赖关系,随着大蒜素浓度的升高其对血管平滑肌细胞增殖的抑制作用也逐渐增加,在24h时,10.00g/L的大蒜素对血管平滑肌细胞增殖的抑制率为13.46%。结论大蒜素可抑制血管紧张素II诱导兔血管平滑肌细胞的增殖,并存在一定的量效依赖关系及时间反应性。
Objective To observe the inhibitory effect of allicin on the proliferation of vascular smooth muscle cells induced by angiotensin II. Methods The cultured rabbit vascular smooth muscle cells were used 3H-TdR incorporation method to observe the effect of allicin on DNA synthesis of vascular smooth muscle cells and the time effect during the proliferation of VSMC stimulated by angiotensin II. RESULTS: Angiotensin II promoted DNA synthesis in quiescent rabbit vascular smooth muscle cells. DNA synthesis reached a peak at 36 h. Allicin significantly inhibited the proliferation of vascular smooth muscle cells induced by angiotensin II and showed a concentration-dependent relationship. With the increase of allicin concentration, it inhibited the proliferation of vascular smooth muscle cells. At 24 hours, the inhibition rate of 10.00 g/L allicin on the proliferation of vascular smooth muscle cells was 13.46%. Conclusion Allicin can inhibit the proliferation of rabbit vascular smooth muscle cells induced by angiotensin II, and there is a dose-effect relationship and time-response.
