目的:观察补肾通脉方对多囊卵巢综合征(polycystic ovarian syndrome,PCOS)胰岛素抵抗(insulin resistance,IR)的大鼠肝脏、脂肪、肌肉和卵巢组织中磷脂酰肌醇-3激酶(PI-3K)p85α亚基的mRNA及蛋白表达的影响。方法:23日龄雌性SD大鼠皮下注射硫酸普拉睾酮钠90 mg.kg-1.d-1,共20 d,联合高脂饮食喂养80 d建立IR的PCOS大鼠模型,成模大鼠随机分为模型组和治疗组,另设15只正常对照组。治疗组大鼠以补肾通脉方进行干预。光镜下观察大鼠排卵情况;葡萄糖氧化酶法测定空腹血糖(FBG);放射免疫法测定空腹血清胰岛素(Fins);RT-PCR检测PI-3K p85α的mRNA表达;免疫组化检测卵巢PI-3K p85α的蛋白表达。结果:与模型组比较,治疗组平均排卵数及排卵率明显增加(P<0.01),Fins浓度显著降低(P<0.01)。治疗组靶组织PI-3K p85α的mRNA和蛋白表达水平均较模型组明显增加(P<0.01或P<0.05)。结论:补肾通脉方具有改善IR和排卵障碍的作用,其机制可能与其上调伴IR的PCOS大鼠靶组织PI-3K p85α的mRNA和蛋白表达有关。
Objective: To observe the effect of Bushen Tongmai Recipe on phosphatidylinositol 3-kinase in liver, fat, muscle and ovary of rats with polycystic ovarian syndrome (PCOS) insulin resistance (IR). 3K) Effects of p85α subunit mRNA and protein expression. METHODS: 23 days old female SD rats were injected subcutaneously with testosterone sulfate 90 mg.kg-1.d-1 for 20 days, and a high-fat diet fed 80 days to establish IR PCOS rat model. Randomly divided into model group and treatment group, and another 15 normal control groups. The rats in the treatment group were treated with Bushentongmai. The ovulation of rats was observed under light microscope; fasting blood glucose (FBG) was determined by glucose oxidase method; fasting serum insulin (Fins) was measured by radioimmunoassay; PI-3K p85α mRNA expression was detected by RT-PCR; ovarian PI- was detected by immunohistochemistry. 3K p85α protein expression. Results: Compared with the model group, the average ovulation number and ovulation rate in the treatment group increased significantly (P<0.01), and the Fins concentration decreased significantly (P<0.01). The mRNA and protein expression levels of PI-3K p85α in the target group were significantly higher than those in the model group (P<0.01 or P<0.05). Conclusion: Bushen Tongmai Decoction can improve the IR and ovulation dysfunction, and its mechanism may be related to the up-regulation of PI-3K p85α mRNA and protein expression in the target tissue of IR rats with PCOS.
