目的检测骨髓基质细胞条件培养液(N-CM)中诱导神经干细胞(NSCs)向神经元方向分化的细胞因子,探讨骨髓基质细胞(BMSCs)调节NSCs分化的机制。方法将收集到的N-CM混匀后经超滤浓缩分为大于5kD和小于5kD两部分,用这两部分培养液分别培养NSCs,确定其中能促进NSCs分化为高比例神经元的部分并进行大鼠蛋白质微阵列检测。结果N-CM分子量大于5kD的部分可以促进NSCs分化为高比例神经元,并通过蛋白质微阵列检测,与单纯骨髓基质培养液相比,有7种细胞因子的表达量上调了1.5倍,分别是CINC-3、CNTF、IFN-γ、IL-1α、MCP-1、TIMP-1和VEGF。结论BMSCs分泌的可溶性分子对NSCs分化为神经元有促进作用,其中某些分子量大于5KD的细胞因子可能在这一调节作用中发挥效应。
Objective To detect the differentiation of neural stem cells (NSCs) into neurons in bone marrow stromal cell conditioned medium (N-CM) and to explore the mechanism of bone marrow stromal cells (BMSCs) regulating the differentiation of NSCs. Methods The collected N-CM was concentrated by ultrafiltration into more than 5kD and less than 5kD. The two mediums were used to culture NSCs respectively, and the part which promoted the differentiation of NSCs into high proportion of neurons was determined and carried out Rat protein microarray detection. Results The fraction of N-CM with molecular weight greater than 5 kD promoted the differentiation of NSCs into high proportion of neurons and detected by protein microarray. Compared with the simple marrow stromal medium, the expression of seven cytokines was up-regulated by 1.5 times CINC-3, CNTF, IFN-γ, IL-1α, MCP-1, TIMP-1 and VEGF. Conclusion Soluble molecules secreted by BMSCs may promote the differentiation of NSCs into neurons. Some cytokines with a molecular weight greater than 5 kD may play an important role in this regulation.
