目的:研究地塞米松对面神经损伤后面神经核小胶质细胞MHCⅠ,Ⅱ类抗原表达的影响.方法:采用免疫组织化学方法,以单克隆抗体OX18检测MHCⅠ类抗原,用单克隆抗体Ox6检测MHCⅡ类抗原,比较地塞米松治疗组与对照组MHCⅠ,Ⅱ类抗原阳性细胞数的变化.结果:地塞米松治疗组损伤面神经核中MHCⅡ类抗原表达的阳性细胞数与对照组相比明显减少(P<0.05),而MHCⅠ类抗原表达两组之间则没有明显改变(P>0.05).结论:地塞米松能下调损伤面神经核中MHCⅡ类抗原的表达,而对MHCⅠ类抗原表达无影响.
Objective: To study the effect of dexamethasone on expression of MHC class I and class II antigens in facial nerve microglia after facial nerve injury. Methods: Immunohistochemical method was used to detect MHC class I antigen with monoclonal antibody OX18. MHC class II antigen was detected with monoclonal antibody Ox6. The changes of MHC class I and class II antigen positive cells in dexamethasone group and control group were compared. Results: Compared with the control group, the number of MHC Ⅱ antigen-positive cells in the denervated facial nerve nucleus of dexamethasone treated group was significantly decreased (P <0.05), while there was no significant difference in the expression of MHC class I antigen between the two groups (P> 0.05). Conclusion: Dexamethasone can down-regulate the expression of MHC class Ⅱ antigen in the injured facial nucleus, but has no effect on the expression of MHC class Ⅰ antigen.
