目的观察胞外三磷酸腺苷(ATP)对体外培养许旺细胞释放谷氨酸(Glu)的影响,并对其作用机制进行探讨。方法用反相高效液相色谱(HPLC)——荧光检测法,分别测定ATP、腺苷和苏拉明对体外培养许旺细胞(SC)释放Glu的影响。结果细胞外ATP可显著促进SC释放Glu;撤除ATP后,Glu浓度迅速回降至正常水平;腺苷不能诱导SC释放Glu;苏拉明可阻止ATP对SC的促Glu释放作用,且此作用可在苏拉明停用后为ATP所逆转。结论细胞外ATP可显著促进SC释放Glu;其作用途径经由P_2型嘌呤能受体所实现。ATP参与外周神经损伤与再生的双向凋节作用。
Objective To observe the effect of extracellular ATP on the release of glutamic acid (Glu) from Schwann cells cultured in vitro and its mechanism of action. Methods The effects of ATP, adenosine and suramin on the release of Glu from Schwann cells cultured in vitro were determined by reversed-phase high-performance liquid chromatography (HPLC) and fluorescence detection. Results Extracellular ATP significantly promoted the release of Glu from SC. After the removal of ATP, the concentration of Glu decreased rapidly back to normal level. Adenosine did not induce SC release of Glu. Suramin blocked the Glu-releasing effect of ATP on SC. Reversal of ATP after withdrawal of suramin. Conclusion Extracellular ATP can significantly promote the release of Glu from SC. The mechanism of action is via P2 purinergic receptor. ATP participates in bidirectional apoptosis of peripheral nerve injury and regeneration.
