目的建立辅酶A中残留溶剂丙酮、巯基乙醇的含量测定方法。方法采用顶空毛细管气相色谱法,HP-INNOWAX毛细管柱(30 m×0.53 mm×1μm),FID检测器。结果丙酮、巯基乙醇达到了完全分离,浓度与峰面积呈良好的线性关系(r>0.999),平均回收率分别为101.29%(RSD=1.32%,n=9)、98.67%(RSD=1.71%,n=9),检测限分别为2.52μg.mL-1、3.95μg.mL-1,样品中丙酮含量为0.06%~4.33%,巯基乙醇含量为0.14%~0.72%。结论本方法快速准确可靠,简便易行,可用于辅酶A的质量控制。
Objective To establish a method for the determination of residual solvents acetone and mercaptoethanol in Coenzyme A Methods Headspace capillary gas chromatography, HP-INNOWAX capillary column (30 m × 0.53 mm × 1 μm), FID detector. The results showed that acetone and mercaptoethanol were completely separated and the linear relationship was obtained with the peak area (r> 0.999). The average recoveries were 101.29% (RSD = 1.32%, n = 9) and 98.67% (RSD = 1.71% , n = 9). The detection limits were 2.52μg.mL-1, 3.95μg.mL-1 respectively. The acetone content in the sample ranged from 0.06% to 4.33% and the content of mercaptoethanol from 0.14% to 0.72%. Conclusion The method is rapid, accurate and reliable, simple and easy to use and can be used for the quality control of Coenzyme A.
