目的观察外源性生长激素(GH)和生长抑素(SS)对人结肠癌细胞株HT-29裸鼠移植瘤模型肿瘤生长和裸鼠血清蛋白水平的影响。方法接种结肠癌细胞株HT-29建立裸鼠移植瘤模型。比较移植肿瘤体积、细胞周期分布、增殖指数(PI)、凋亡率、bcl-2和bax mRNA水平,以及裸鼠血清蛋白水平。结果生长抑素能够明显抑制移植瘤的生长(P<0.05)、降低移植瘤PI(P<0.05)、促进移植瘤细胞凋亡(P<0.01)、降低移植瘤bcl-2 mRNA表达(P<0.05)、提高移植瘤bax mRNA表达(P<0.05),生长激素则表现出相反的作用。生长激素能够改善移植瘤导致的低蛋白血症(P<0.05)。结论外源性生长抑素能够显著抑制人结肠癌裸鼠移植瘤生长,外源性生长激素单独或联合生长抑素应用能够改善荷人结肠癌移植瘤裸鼠低蛋白血症,但具有潜在的促进肿瘤生长的作用。
Objective To observe the effect of exogenous growth hormone (GH) and somatostatin (SS) on the growth of human colon cancer cell line HT-29 xenograft in nude mice and serum protein level in nude mice. Methods The colon cancer cell line HT-29 was inoculated to establish a nude mouse xenograft model. Tumor volume, cell cycle distribution, proliferation index (PI), apoptosis rate, bcl-2 and bax mRNA levels, and serum protein levels in nude mice were compared. Results Somatostatin could significantly inhibit the growth of xenografts (P <0.05), reduce the PI (P <0.05) and promote the apoptosis of xenografts (P <0.01), and decrease the expression of bcl- 2 mRNA (P <0.05), and increased the expression of bax mRNA (P <0.05) in tumor xenografts. However, growth hormone had the opposite effect. Hormone can improve tumor-induced hypoproteinemia (P <0.05). Conclusion Exogenous somatostatin can significantly inhibit the growth of transplanted human colon cancer xenografts in nude mice. The application of exogenous growth hormone alone or in combination with somatostatin can improve the hypoproteinemia in nude mice bearing human colon carcinoma xenografts.
