目的:研究十二味穿甲片对大鼠免疫性肝纤维化模型的影响,并探讨其相关的作用机制。方法:将雄性SD大鼠随机分为正常组,模型组,阳性药组(秋水仙碱,0.1 mg·kg-1),中药十二味穿甲片2.8,1.4,0.7 g·kg-1剂量组。除正常组外,其余各组采用猪血清制备大鼠免疫性肝纤维化模型,造模同时给药进行抗肝纤维化干预,共6周。通过计算体重、肝脏指数,检测肝功能丙氨酸转氨酶(ALT),天冬氨酸转氨酶(AST),总蛋白(TP),白蛋白(ALB),球蛋白(GLO)水平和白球比(A/G),酶联免疫吸附测定(ELISA)法检测血清透明质酸(HA)含量。苏木素-伊红(HE)染色以观察肝组织病理变化及马松(Masson)三色染色观察胶原纤维增生情况,评价药物的抗纤维化作用。结果:与正常组比较,模型组大鼠血清ALT,HA水平明显升高,肝指数明显升高(P<0.01),ALB,A/G明显降低(P<0.01),模型组肝组织的病变较明显;与模型组比较,十二味穿甲片除对血清AST,GLO的变化无明显影响外,能够显著降低肝纤维化大鼠血清ALT和纤维化指标HA水平(P<0.01),不同程度地升高血清TP,ALB和A/G,并能显著地减轻肝组织的病变,尤以十二味穿甲片2.8 g·kg-1剂量效果最显著。结论:十二味穿甲片可明显改善模型大鼠肝纤维化程度,显示出一定的抗肝纤维化作用。
Objective: To study the effects of 12-flavonoid-penetrating decoction on immune-induced hepatic fibrosis in rats and its related mechanism. Results: Compared with normal group, serum ALT and HA in model group were significantly increased, liver index was significantly increased (P <0.01), ALB and A / G were significantly decreased (P <0.01) Compared with the model group, the mean value of serum ALT and fibrosis indexes of liver fibrosis rats was significantly lower than that of the model group (P <0.01) The level of serum TP, ALB and A / G were increased to a certain degree and the pathological changes of liver tissue were significantly alleviated. Especially the dose of 2.8 g · kg-1 was the most significant. Conclusion: Twelve flavors and puncture tablets can obviously improve the degree of hepatic fibrosis in model rats, and show a certain anti-hepatic fibrosis effect.
