采用SCoT分子标记分析4种黄连属药用植物的遗传多样性和亲缘关系,并用POPGENE计算遗传参数,TREECONW软件进行聚类分析。结果表明:28条引物共检测到434条带,其中多态性条带430条;各遗传参数均表明黄连属药用植物种间的遗传多样性极为丰富(种间PPB=99.1%,N_a=1.990 6,N_e=1.329 3,H=0.212 2,I=0.337 8),但种内遗传多样性较低(种内PPB=16.8%,N_a=1.168 2,N_e=1.073 0,H=0.043 7,I=0.067 7);遗传分化系数G_(st)=0.794 0,说明79.4%的遗传变异来源于种间;同种黄连属药用植物的不同植株均聚在一起,证实SCoT分子标记可作为黄连属药用植物亲缘关系研究的有效方法之一。
SCoT molecular markers were used to analyze the genetic diversity and genetic relationship of four species of Coptis, and genetic parameters were calculated by POPGENE. The TREECONW software was used for cluster analysis. The results showed that a total of 434 bands were detected with 28 primers, of which 430 bands were polymorphic. The genetic parameters indicated that the genetic diversity of Medicinal plants of Coptis was extremely rich (inter-species PPB = 99.1%, N_a = 1.990 6, N_e = 1.329 3, H = 0.212 2, I = 0.337 8), but the intraspecific genetic diversity was low (intraspecific PPB = 16.8%, N_a = 1.168 2, N_e = 1.073 0, H = 0.043 7, I = 0.067 7). The genetic differentiation coefficient G st (st) was 0.794 0, indicating that 79.4% of the genetic variation originated from interspecific crosses. Different plants belonging to the same genus Coptis were clustered together and confirmed that the SCoT It is one of the effective methods to study the genetic relationship of medicinal plants.
