目的 :探讨微囊化癌胚抗原 (CEA)转基因细胞疫苗免疫小鼠后 ,对小鼠免疫功能的影响。方法 :采用 3H- Td R掺入法 ,测定 Con A诱导荷瘤小鼠脾淋巴细胞的增殖作用 ;采用 3H- Td R后标记法 ,测定荷瘤小鼠脾淋巴细胞 NK细胞毒 ;以 EL ISA法测定小鼠脾淋巴细胞 IL- 2和 IFNγ细胞因子水平 ;利用流式细胞术检测 CEA疫苗免疫对小鼠脾 T淋巴细胞亚群 CD3、 CD4、 CD8百分率 ,CD4 /CD8比值和 NK细胞的影响 ;采用 RT- PCR方法检测 IL - 2、 IFNγ m RNA的表达水平。结果 :微囊化 CEA疫苗免疫可以有效增强 Con A诱导的小鼠淋巴细胞增殖能力 ,促进 IL- 2、IFNγ的产生以及增强 NK细胞的细胞毒活性 (P<0 .0 5 ,P<0 .0 1,P<0 .0 5 ) ;微囊化 CEA疫苗能提高荷瘤小鼠 CD4 /CD8比值及 NK细胞数 (P<0 .0 5 ,P<0 .0 5 ) ;促进脾细胞中细胞因子 IL - 2、 INFγ m RNA的表达。结论 :微囊化 CEA转基因细胞疫苗具有增强荷瘤小鼠免疫功能的作用 ,提示该疫苗可作为有效的抗 CEA阳性肿瘤的治疗性疫苗。
Objective: To investigate the effect of microencapsulated carcinoembryonic antigen (CEA) transgenic mice vaccinated on immune function in mice. Methods: 3H-TdR incorporation assay was used to determine the proliferation of splenic lymphocytes induced by Con A in tumor-bearing mice. The cytotoxicity of NK cells in spleen lymphocytes was detected by 3H-TdR labeling. Method was used to determine the levels of IL-2 and IFNγ in mouse splenic lymphocytes. The effects of CEA vaccine on the percentage of CD3, CD4, CD8, CD4 / CD8 and NK cells in splenic T lymphocyte subsets were detected by flow cytometry The expression of IL-2 and IFNγ m RNA was detected by RT-PCR. Results: Immunization with microencapsulated CEA vaccine could effectively enhance the lymphocyte proliferation induced by Con A, promote the production of IL-2 and IFNγ and enhance the cytotoxic activity of NK cells (P <0.05, P <0. 0 1, P <0. 05). The microencapsulated CEA vaccine increased the ratio of CD4 / CD8 and NK cells in tumor-bearing mice (P <0.05, P <0.05) Expression of cytokines IL - 2 and INFγ m RNA. CONCLUSION: The microencapsulated CEA transgenic cell vaccine has the effect of enhancing the immune function of tumor-bearing mice, suggesting that the vaccine can be used as an effective therapeutic vaccine against CEA-positive tumors.
