采用共振Ravleigh散射并结合平衡透析法研究了I~-与人血清白蛋白(humanserum albumin,简称HAS)或牛血清白蛋白(Bovine Serum Album in,简称BSA)结合平衡.观测到I~-对HSA和BSA的共振Ragleigh散射、倍频散射有增强效应,却导致荧光猝灭.平衡透析结果表明: I~-与血清白蛋白的结合平衡不符合Scatchard模型,却能较好地符合相平衡分配规律,表观相分配常数受pH影响,其数量级为104.Cl-和其他阴离子很可能是通过改变溶液的离子强度影响I-的结合.根据不同的pH条件下的实验结果,推测I-是与白蛋白上质子化的碱性氨基酸残基结合.
The binding equilibrium between I ~ - and human serum albumin (HAS) or bovine serum albumin (BSA) was investigated by resonance Ravleigh scattering combined with equilibrium dialysis. Observed I ~ - Resonance Ragusigh scattering and frequency doubling scattering of HSA and BSA have an enhancement effect, but lead to fluorescence quenching. The equilibrium dialysis results showed that the binding equilibrium of I ~ - and serum albumin did not fit the Scatchard model, but it was in good agreement with the law of phase equilibrium partition, and the apparent phase partition constant was affected by pH, with an order of magnitude of 104. Cl- and other anions It is likely that I-binding is affected by changing the ionic strength of the solution. According to the experimental results under different pH conditions, it is presumed that I- is bound to a protonated basic amino acid residue on albumin.
