目的 研究螺旋藻多糖中的均一组分SPPA 1的分离方法、糖组成、分子量及生物活性。方法 采用丙酮分部沉淀 ,三氯乙酸去蛋白 ,再经SephadexG 75和CM SephadexC 5 0柱色谱得到SPPA 1,用气相色谱法测定单糖组成 ,GPC法测定分子量 ,IR和NMR法确定糖苷键类型。结果 经高压液相色谱和毛细管电泳鉴定为均一性组分。其总糖含量为 91 70 % ,N含量为 0 96 % ,分子量为 6 9 0 0× 10 4 Da。糖组成分析表明其由单一的葡萄糖组成 ,红外光谱、核磁共振1HNMR谱分析表明 ,其单糖类型为α 吡喃型。ESR研究表明SPPA 1具有清除O·2 自由基活性。结论 SPPA 1为一具抗氧化活性的葡聚糖
Objective To study the separation method, sugar composition, molecular weight and biological activity of SPPA 1, a homogeneous component in Spirulina platensis polysaccharides. Methods Acetone was used to precipitate and protein was deproteinized by trichloroacetic acid. SPPA was obtained by column chromatography on Sephadex G 75 and CM Sephadex C 50. The monosaccharide composition was determined by gas chromatography, the molecular weight was determined by GPC, and the glycosidic bond was determined by IR and NMR. . Results The homogenous components were identified by high pressure liquid chromatography and capillary electrophoresis. Its total sugar content is 91 70 %, its N content is 0 96 %, and its molecular weight is 6 9 0 × 10 4 Da. Analysis of the sugar composition showed that it consisted of a single glucose. IR and 1H NMR spectroscopy showed that the monosaccharide type was α-pyran. ESR studies showed that SPPA 1 has O.2 free radical activity. Conclusion SPPA 1 is a glucan with antioxidant activity
