目的:研究女贞子提取物(Ligustrum lucidum Ait Extract,LLAE)对链脲佐菌素(streptozocin,STZ)诱导的糖尿病肾病(DN)大鼠模型肾皮质血管内皮生成因子(VEGF)mRNA表达水平的影响,探讨女贞子提取物对肾脏的保护作用机制。方法:将65只SD大鼠随机分为正常对照组(N组),糖尿病肾病模型组(M组),女贞子提取物低(LLLAE,0.2 g/kg·d)、中(MLLAE,0.4 g/kg·d)、高剂量(HLLAE,0.8 g/kg·d)组,缬沙坦阳性对照组(V,10 mg/kg·d)。模型组、女贞子提取物低、中、高剂量组、缬沙坦组大鼠用链脲佐菌素(STZ)按55 mg·kg~(-1)空腹腹腔注射法构建糖尿病肾病大鼠模型。各实验组从造模成功后开始给药。实验第8周末处死大鼠,检测大鼠体重(BW)、肾脏肥大指数(KW.BW~(-1))、空腹血糖(FBG)、血尿素氮(Bun)、血清肌酐(Scr)和24 h尿白蛋白定量;HE染色法观察大鼠肾脏病理学变化;酶联免疫法和逆转录聚合酶链反应(RT-PCR)分别检测肾皮质VEGF蛋白含量和VEGFmRNA的表达。结果:女贞子提取物0.4、0.8 g·kg~(-1)剂量组与模型组相比体重明显上升,FBG、Bun、Scr、24 h尿蛋白定量、VEGF蛋白和VEGFmRNA表达显著降低,差别有非常显著意义(P<0.05或P<0.01)。结论:女贞子提取物可减少DN大鼠的尿蛋白排泄量、改善肾脏功能、延缓DN进程,其机制可能与通过降糖作用抑制了肾皮质内VEGFmRNA的表达水平,从而减少了VEGF的生成有关。

Conclusion: Fructus Ligustri Lucidi extract can reduce urinary protein excretion, improve renal function and delay DN progression in DN rats. The mechanism may be related to reducing the expression of VEGF mRNA in the renal cortex via hypoglycemic effect related.
