目的:研究丹龙醒脑方对脑缺血再灌注大鼠血管新生及碱性成纤维细胞生长因子(b FGF)的影响。方法:用线栓法制备大鼠大脑中动脉栓塞缺血再灌注(MCAO)模型,2 h后开放血流再灌注。大鼠造模成功后分为6组,丹龙醒脑方大(14.76 g/kg)、中(7.38 g/kg)、小(3.69 g/kg)剂量组,尼莫地平(10.80 mg/kg)组,模型组,假手术组,灌胃7 d后处死大鼠取脑,用免疫组化法测定缺血海马区的内皮细胞数和微血管密度(MVD)及bFGF的表达。结果:与假手术组比较,模型组大鼠内皮细胞计数及微血管密度、bFGF蛋白表达均增强(P<0.05或P<0.01);与模型组比较,丹龙醒脑方组内皮细胞数、微血管密度及bFGF蛋白表达均有明显升高,差异有统计学意义(P<0.05)。结论:丹龙醒脑方能够促进脑缺血再灌注大鼠缺血脑组织血管新生,对缺血脑组织具有保护作用,其作用机制可能与上调bFGF蛋白表达有关。
Objective: To study the effect of Danlongxingnao Prescription on angiogenesis and basic fibroblast growth factor (b FGF) in rats with cerebral ischemia / reperfusion. Methods: MCAO model was established by middle cerebral artery occlusion (MCAO). After 2 hours, blood flow and reperfusion were performed. Rats were divided into 6 groups: Dulong Xingnao Fangfang (14.76 g / kg), middle (7.38 g / kg) and small (3.69 g / kg) The rats in the model group and the sham operation group were sacrificed 7 days after gavage, and the brain was removed. The number of endothelial cells and microvessel density (MVD) and the expression of bFGF in the hippocampus were determined by immunohistochemistry. Results: Compared with the sham operation group, the endothelial cell count, microvessel density and bFGF protein expression in the model group were significantly increased (P <0.05 or P <0.01). Compared with the model group, the number of endothelial cells, microvessel Density and bFGF protein expression were significantly increased, the difference was statistically significant (P <0.05). CONCLUSION: Danlongxingnao Decoction can promote angiogenesis in ischemic brain tissue of rats with cerebral ischemia-reperfusion, which has a protective effect on ischemic brain tissue. Its mechanism may be related to the up-regulation of bFGF protein expression.
