目的探索豚鼠内耳Hensen细胞(Hensen cells,HC)中透明物质的性质及分布。方法 10只豚鼠分离制备好耳蜗单离Hensen细胞后,采用中性脂滴的特异分子探针Bodipy 493/503、苏丹Ⅲ、油红O染色及锇酸再固定四种染色方法对内耳组织进行染色,观察耳蜗不同区域Hensen细胞中脂滴的分布情况。结果豚鼠耳蜗不同区域Hensen细胞内均存在高透亮物质,从顶回至底回其体积逐渐变小,数量逐渐增加,至底回逐渐消失;该物质经Bodipy 493/503分子探针特异标记为绿色,被苏丹Ⅲ染色为橘红色,被油红O染色为宝石红,被锇酸后固定后呈黑色颗粒,证实为脂滴。结论四种染色方法对内耳组织的染色结果均提示内耳Hensen细胞中透明物质为脂类物质。
Objective To explore the properties and distribution of transparent substances in Hensen cells (HC) of guinea pig inner ear. Methods Ten human guinea pigs were isolated and prepared into Hensen cells. The inner ear tissue was stained with Bodipy 493/503, Sudan Ⅲ, Oil Red O and Osmium Osate, respectively. , Observe the distribution of lipid droplets in Hensen cells in different regions of the cochlea. Results High translucent material was found in all Hensen cells in different regions of guinea pig cochlea. The volume gradually increased from the top to the bottom and gradually disappeared. In the end, the substance gradually disappeared. The substance was specifically labeled as green by the Bodipy 493/503 molecular probe , Was dyed by Sudan Ⅲ orange red, oil red O stained ruby red, osmium acid was fixed after the black particles, confirmed as lipid droplets. Conclusion The staining results of four kinds of staining methods on the inner ear tissues suggest that the transparent substance in the inner ear Hensen cells is a lipid substance.
