目的探讨凋亡蛋白抑制因子Survivin及其剪接变异体SurvivinΔex3、Survivin2B在脑胶质瘤发生发展中的作用。方法采用逆转录聚合酶链反应(RTPCR)检测Survivin及其剪接变异体SurvivinΔex3、Survivin2BmRNA在62例脑胶质瘤和10例正常脑组织标本中的表达水平。结果Survivin、SurvivinΔex3及Survivin2BmRNA在脑胶质瘤中表达阳性率分别为67.7%、30.6%和17.7%,三者在正常脑组织中均无表达;Survivin和SurvivinΔex3mRNA表达阳性率在脑胶质瘤及正常脑组织之间分别具有极显著差异(P=0.000<0.01)和显著差异(P=0.010<0.05);SurvivinmRNA表达阳性率在低恶性度和高恶性度脑胶质瘤之间以及脑胶质瘤不同病理级别间分别具有极显著差异(P=0.002<0.01,P=0.004<0.01)。结论Survivin在脑胶质瘤发生发展过程中具有重要作用,其剪接变异体SurvivinΔex3、Survivin2B也发挥一定作用;Survivin可作为脑胶质瘤生物治疗的一个良好分子靶点。
Objective To investigate the role of Survivin, a Survivin inhibitor of apoptosis protein, in the development of glioma and its splicing variants SurvivinΔex3 and Survivin2B. Methods The expression of Survivin and SurvivinΔex3 and Survivin2B mRNA in 62 gliomas and 10 normal brain tissues were detected by reverse transcription polymerase chain reaction (RTPCR). Results The positive rates of Survivin, SurvivinΔex3 and Survivin2B mRNA in gliomas were 67.7%, 30.6% and 17.7%, respectively. The positive rates of Survivin and SurvivinΔex3 mRNA in gliomas and normal There was significant difference (P = 0.000 <0.01) and significant difference (P = 0.010 <0.05) between the brain tissues. The positive rate of Survivin mRNA expression in glioma between low and high grade malignant gliomas There were significant differences among different pathological grades (P = 0.002 <0.01, P = 0.004 <0.01). Conclusions Survivin plays an important role in the development of gliomas. Survivin also plays a role in its expression. Survivin can be used as a good molecular target for biological treatment of glioma.
