目的】探讨血管紧张素Ⅱ受体拮抗剂洛沙坦(losartan)在高血压病血管重塑过程中的作用及作用机制。【方法】①应用ACAS570共聚激光扫描显微系统,通过荧光免疫组化方法,对洛沙坦作用下体外培养的大鼠血管平滑肌细胞中的核细胞增殖抗原(PCNA)及与增殖相关的Cmyc基因表达产物进行定量检测。②测定洛沙坦作用下的体外培养的大鼠血管平滑肌细胞的增殖活性(MTT)值。【结果】洛沙坦作用下血管平滑肌细胞的增殖活性(MTT)降低;实验组和对照组分别为02045±00013,02239±00010(P<005)。PCNA及Cmyc的表达都明显降低;实验组和对照组PCNA分别为1338.56±232.56,154270±20004(P<001)。Cmyc分别为152803±12514,174035±20591(P<001)。【结论】洛沙坦通过影响血管平滑肌细胞的增殖活性而参与血管壁结构的改变。
Objective] To investigate the role and mechanism of losartan, an angiotensin Ⅱ receptor antagonist, in the process of vascular remodeling in hypertensive patients. 【Methods】 ① ACAS570 laser scanning microscopy was used to detect the expression of proliferating cell nuclear antigen (PCNA) and proliferation-related Cmyc gene in rat vascular smooth muscle cells cultured with losartan by fluorescence immunohistochemistry The expression product was quantified. ② To determine the proliferation activity (MTT) of rat vascular smooth muscle cells cultured with losartan. [Results] The proliferation activity (MTT) of VSMC decreased in losartan group. The experimental group and the control group were respectively 02045 ± 00013 and 02239 ± 00010 (P <005). PCNA and Cmyc expression were significantly reduced; experimental group and control group PCNA were 1338.56 ± 232.56,154270 ± 20004 (P <001). Cmyc were 152803 ± 12514,174035 ± 20591 (P <001). 【Conclusion】 Losartan is involved in the structural changes of vascular wall by affecting the proliferation activity of vascular smooth muscle cells.
