目的 :为研究细胞色素P4 50同工酶CYP2D6在人群中的代谢多态性 ,本文建立了人尿中右美沙芬 (DM)及其O 去甲基代谢物 3 羟基 N 甲基 吗喃 (DT)的气相色谱分析方法。方法 :尿样经提取后 ,以HP 1毛细管柱作为分离柱 ,FID为检测器 ,正二十二烷为内标进行气相色谱分析。结果 :在此实验条件下 ,右美沙芬、内标物和代谢物的保留时间分别为 8.5,9.9和 1 1 .2min ;尿样中DM在 0 .1 0~ 2 .0 μg/ml、DT在 0 .2 0~ 2 0 .0 0 μg/ml范围内线性关系良好 ,回收率分别 95.7%~ 1 0 4 .4 % (DM )和 93 .3 %~ 98.3 % (DT) ;样品测定的RSD分别为 2 .5%和 3 .1 %。结论 :该法简便可靠 ,能满足CYP2D6代谢多态性研究的要求。
OBJECTIVE: To investigate the metabolic polymorphism of CYP2D6, a cytochrome P450 isoenzyme, in human urine, a method was developed for the determination of dextromethorphan (DM) and its demethylation product 3-hydroxy-N-methylmorpholine ) Gas chromatography method. Methods: After the urine samples were extracted, HP 1 capillary column was used as separation column, FID was used as detector, and n-docosane was used as internal standard for gas chromatographic analysis. Results: Under these experimental conditions, the retention time of dextromethorphan, internal standard and metabolites were 8.5, 9.9 and 11.2 min respectively; the DM in the urine sample ranged from 0.001 to 2.0 μg / ml and DT The calibration curve was linear over the range of 0.2-2.0.0 μg / ml, recoveries ranged from 95.7% to 104.4% (DM) and from 93.3% to 98.3% (DT), respectively. RSD was 2.5% and 3.1% respectively. Conclusion: The method is simple and reliable, and can meet the CYP2D6 metabolic polymorphism research requirements.
