目的:探讨脑梗塞后给予大鼠颈淋巴管阻塞对缺血侧大脑皮质基质金属蛋白酶9表达水平及血脑屏障通透性和脑含水量等的影响。方法:用线栓法复制大鼠大脑中动脉阻塞(MCAO)模型,以及行大鼠大脑中动脉阻塞15min后摘除双侧颌下腺的颈浅和颈深淋巴结,造成脑梗塞后颈淋巴管阻塞(MCAO+CLB)模型去检测缺血侧大脑皮质脑含水量、血脑屏障通透性和基质金属蛋白酶9mRNA表达水平和免疫活性的变化。结果:在不同的时点,上述各指标在MCAO+CLB组大鼠明显高于MCAO组。结论:MCAO后,CLB通过上调梗塞侧皮质MMP-9的表达水平而加重脑水肿和提高血脑屏障通透性。
OBJECTIVE: To investigate the effect of cervical lymphatic obstruction on the expression of matrix metalloproteinase 9 (MMP-9), permeability of brain-blood-brain barrier (BBB) and brain water content in rat cerebral cortex after cerebral infarction. Methods: The middle cerebral artery occlusion (MCAO) model was induced by thread occlusion and the neck superficial and deep cervical lymph nodes of the bilateral submandibular glands were removed 15 min after the middle cerebral artery occlusion + CLB) model was used to detect changes of brain water content, blood-brain barrier permeability and matrix metalloproteinase 9 mRNA expression and immunological activity in ischemic cortex. Results: At different time points, the above indexes were significantly higher in MCAO + CLB group than MCAO group. Conclusion: After MCAO, CLB aggravates cerebral edema and enhances the permeability of the blood-brain barrier by up-regulating the expression of MMP-9 in infarcted cortex.
