目的检测乙醛脱氢酶1(ALDH1)mRNA在卵巢癌中的表达水平,探讨其用于卵巢癌辅助诊断的临床意义。方法选取61例卵巢癌组织、57例卵巢良性组织和38例卵巢正常组织标本,提取总RNA,逆转录-聚合酶链式反应(RT-PCR)方法检测其ALDH1 mRNA表达水平,并分析其与临床病理特征之间的相关性。结果 ALDH1在卵巢癌中表达阳性率为52.46%,显著高于卵巢良性组织(29.82%)和正常组织(10.53%),差异有统计学意义(P<0.01);在卵巢癌中,其mRNA表达水平随着病理分级的增高而增高,3期、4期明显高于1期、2期;淋巴结转移组表达量高于未转移组,差异有统计学意义(P<0.05);卵巢癌ALDH1 mRNA表达量与肿瘤患者年龄无明显相关性。结论 ALDH1在卵巢癌中高表达,可能作为卵巢癌辅助诊断指标之一,用来判断卵巢癌分期、分级及淋巴结转移情况。
Objective To detect the expression of aldehyde dehydrogenase 1 (ALDH1) mRNA in ovarian cancer and to explore its clinical significance in the diagnosis of ovarian cancer. Methods 61 cases of ovarian cancer tissues, 57 cases of ovarian benign tissues and 38 cases of normal ovarian tissues were collected. The expression of ALDH1 mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR) Correlation between clinicopathological features. Results The positive expression rate of ALDH1 in ovarian cancer was 52.46%, which was significantly higher than that in benign ovarian tissues (29.82%) and normal tissues (10.53%) (P <0.01). Conclusions ALDH1 is overexpressed in ovarian cancer and may be used as a diagnostic marker for ovarian cancer to determine the stage, grade and lymph node metastasis of ovarian cancer.
