目的:建立同时测定丹参中二氢丹参酮Ⅰ、降鼠尾草氧化物、隐丹参酮、丹参酮Ⅰ、丹参酮ⅡA5种脂溶性成分的超高效液相色谱(UPLC)分析方法。方法:采用Waters Acquity UPLC系统,Acquity UPLC BEH C18(50mm×2.1mm,1.7μm)色谱柱,流动相为乙腈-水,以流速为0.5mL·min-1进行梯度洗脱;柱温:35℃;样品经甲醇超声提取后,用UPLC-PDA进行分析检测,检测波长为280nm。结果:二氢丹参酮Ⅰ、降鼠尾草氧化物、隐丹参酮、丹参酮Ⅰ、丹参酮ⅡA5种被测成分在线性范围内均具有良好的线性关系(r≥0.9997);平均回收率在98.5%~103.1%之间,RSD≤4.2%。结论:UPLC分离效果及重复性好,且快速、简便,可作为丹参的质量控制方法。
OBJECTIVE: To establish a method for the simultaneous determination of five lipid-soluble constituents of Salvia miltiorrhiza Bge in Salvia miltiorrhiza I, Salvia mollis, Cryptotanshinone, Tanshinone I and Tanshinone IIA by ultra performance liquid chromatography (UPLC). Methods: A Waters Acquity UPLC system was used on a Acquity UPLC BEH C18 (50 mm × 2.1 mm, 1.7 μm) column with a mobile phase of acetonitrile-water and gradient elution at a flow rate of 0.5 mL · min -1. The column temperature was 35 ℃ The sample was extracted by methanol and analyzed by UPLC-PDA. The detection wavelength was 280nm. Results: There was a good linear relationship between dihydrotanshinone Ⅰ, salbutamol, cryptotanshinone, tanshinone Ⅰ and tanshinone ⅡA in the linear range (r ≥ 0.997). The average recoveries ranged from 98.5% to 103.1 %, RSD ≤ 4.2%. Conclusion: UPLC separation efficiency and repeatability, and fast, simple and can be used as a quality control method of Salvia miltiorrhiza.
