目的探讨螺内酯对单侧输尿管梗阻(UUO)大鼠肾间质纤维化的影响。方法2004-12~2005-03对UUO大鼠给予螺内酯20mg·kg-1·d-1及50mg·kg-1·d-1灌胃,观察第14天血清钾、钠、肌酐、血压的变化及肾间质纤维化程度、转化生长因子-β1(TGF-β1)、α-平滑肌肌动蛋白(α-SMA),纤维连接蛋白(FN)的表达。结果与UUO组相比,两治疗组血清钾、钠、肌酐、血压差异均无显著性(P>0.05),肾小管间质纤维化程度,TGF-β1、α-SMA、FN的表达与UUO组差异有显著性(P<0.05),两治疗组之间差异无显著性(P>0.05)。结论螺内酯可通过抑制细胞因子释放和肾小管-间质细胞的表型转化等非血压依赖机制延缓肾间质纤维化。
Objective To investigate the effect of spironolactone on renal interstitial fibrosis in rats with unilateral ureteral obstruction (UUO). METHODS: Rats in UUO group were given intragastric administration of 20 mg · kg-1 · d-1 and 50 mg · kg-1 · d-1 orally from December 2004 to March 2005, and the changes of serum potassium, sodium, creatinine and blood pressure were observed on the 14th day And the degree of renal interstitial fibrosis, the expression of TGF-β1, α-SMA and FN. Results Compared with UUO group, there was no significant difference in serum potassium, sodium, creatinine and blood pressure between the two treatment groups (P> 0.05), the degree of tubulointerstitial fibrosis, the expression of TGF-β1, α-SMA and FN, There was significant difference between the two groups (P <0.05). There was no significant difference between the two groups (P> 0.05). Conclusion Spironolactone can delay renal interstitial fibrosis by inhibiting the release of cytokines and phenotypic transformation of tubulointerstitial cells.
