目的探讨生存素(survivin)、周期蛋白E(cyclin E)和端粒酶相关蛋白(hTP1)在星形细胞胶质瘤中的表达与意义。方法应用免疫组织化学SP法、原位杂交法分别检测106例星形细胞胶质瘤组织中生存素、周期蛋白E和hTP1的表达,分析它们之间的相互关系。结果生存素定位于肿瘤细胞胞浆,周期蛋白E和hTP1主要定位于肿瘤细胞核。生存素、周期蛋白E和hTP1的阳性表达率分别为55.7%(59/106)、61.3%(65/106)和84.9%(90/106)。生存素、周期蛋白E表达和hTP1标记指数均与星形细胞胶质瘤病理分级密切相关(P<0.05)。生存素和周期蛋白E阳性共表达率均为50.9%(54/106),两者的表达呈正相关(P<0.01)。生存素、周期蛋白E表达与hTP1标记指数有相关性(P<0.01)。结论生存素、周期蛋白E和hTP1表达与星形细胞胶质瘤恶性程度有关,其表达异常可能在星形细胞胶质瘤的形成和恶变中发挥重要作用。
Objective To investigate the expression and significance of survivin, cyclin E and telomerase-related protein (hTP1) in astrocytic gliomas. Methods The expressions of survivin, cyclin E and hTP1 in 106 astrocytic glioma tissues were detected by immunohistochemical SP method and in situ hybridization method respectively, and their correlations were analyzed. Results Survivin was located in the cytoplasm of tumor cells. Cyclin E and hTP1 mainly localized in the nucleus of tumor cells. The positive rates of survivin, cyclin E and hTP1 were 55.7% (59/106), 61.3% (65/106) and 84.9% (90/106), respectively. Survivin, cyclin E expression and hTP1 labeling index were closely related to the pathological grade of astrocytic glioma (P <0.05). Survivin and cyclin E positive co-expression rates were 50.9% (54/106), the two were positively correlated (P <0.01). Survivin and cyclin E expression correlated with hTP1 labeling index (P <0.01). Conclusion The expressions of survivin, cyclin E and hTP1 are related to the malignant degree of astrocytic gliomas. The abnormal expression may play an important role in the formation and malignant transformation of astrocytic glioma.
