建立测定小鼠血清中乳酸含量的高效液相色谱方法。血清样品经高氯酸溶液沉淀蛋白质,离心后取清液,采用Eclipse plus C18色谱柱,流动相为5%甲醇水溶液(含0.2%磷酸),流速为0.8mL.min-1,紫外检测波长为210nm,采用外标法进行定量。结果显示乳酸在4.88×10-3—5g.L-1浓度范围内线性关系良好(r=0.9999),检出限为1.5×10-3g.L-1。所建立的方法具有分析速度快、灵敏度高、精密度好等优点。
To establish a method for the determination of lactic acid in mouse serum by high performance liquid chromatography. Serum samples were precipitated protein by perchloric acid solution, centrifuged to take the supernatant, using Eclipse plus C18 column, the mobile phase was 5% aqueous methanol (containing 0.2% phosphoric acid), the flow rate of 0.8mL.min-1, UV detection wavelength 210nm, using external standard method for quantitative. The results showed that the linear range of lactate was 4.88 × 10-3-5g.L-1 (r = 0.9999). The detection limit was 1.5 × 10-3g.L-1. The established method has the advantages of fast analysis, high sensitivity and good precision.
