目的观察阿里红提取物对Aβ_(25-35)诱导老年性痴呆大鼠的学习记忆力和海马乙酰胆碱酯酶活性的影响。方法采用侧脑室注射凝聚态Aβ_(25-35)蛋白诱导老年性痴呆(Alzheimer’s disease,AD)的大鼠模型。取SD大鼠60只,随机分为对照组、模型组(11.5μmol/L Aβ_(25-35))、阿里红提取物低剂量(11.5μmol/L Aβ_(25-35)+0.25 mg/kg阿里红乙酸乙酯部位)组、阿里红提取物中剂量(11.5μmol/LAβ_(25-35)+0.5 mg/kg阿里红乙酸乙酯部位)组、阿里红提取物高剂量(11.5μmol/L Aβ_(25-35)+1 mg/kg阿里红乙酸乙酯部位)组5组。对照组及模型组动物每天固定时间分别给予5μL的生理盐水和Aβ_(25-35),阿里红提取物各剂量组动物分别给予相应剂量的药物。SD大鼠连续给药10 d后,用Morris水迷宫方法对动物进行空间记忆能力测试;采用HE染色法观察各组大鼠神经元病理形态的改变;测定受试动物大脑海马组织乙酰胆碱酯酶(ACh E)含量。结果与模型组比较,阿里红提取物高、中、低剂量组大鼠逃逸潜伏期明显缩短,目标象限停留时间明显延长,穿越平台次数增加,差异有统计学意义(P<0.05);阿里红提取物高、中、低剂量组大鼠海马神经细胞数量明显增多,ACh E活性明显降低。结论阿里红提取物乙酸乙酯部位能改善Aβ_(25-35)诱导所致AD大鼠学习记忆能力障碍,亦能降低脑内乙酰胆碱酯酶活性。
Objective To observe the effects of extract of Alisma orientalis on the learning and memory and the activity of acetylcholinesterase in the hippocampus of Alzheimer ’s disease rats induced by Aβ 25-25. Methods The rat model of Alzheimer’s disease (AD) was induced by intracerebroventricular injection of condensed Aβ_ (25-35) protein. Sixty SD rats were randomly divided into control group, model group (11.5μmol / L Aβ_ (25-35)), low dose of Alirima extract (11.5μmol / L Aβ_ (25-35) +0.25 mg / kg The middle part of Alimentin extract (11.5μmol / LAβ_ (25-35) +0.5 mg / kg alirimary ethyl acetate) group, the high dose of Alirima extract (11.5μmol / L Aβ_ (25-35) +1 mg / kg Ali red ethyl acetate) group of 5 groups. Control group and model group were given fixed daily 5μL of saline and Aβ_ (25-35), Ali red extract of each dose group were given the appropriate dose of the drug. Sprague-Dawley rats were administered with Morris water maze for 10 days. The morphological changes of neurons in each group were observed by HE staining. Results Compared with the model group, the escape latency of the high, middle and low doses of Alisma orientalis significantly shortened, the residence time of the target quadrant significantly prolonged, the number of crossing the platform increased, the difference was statistically significant (P <0.05) The number of hippocampal neurons, the activity of ACh E in the high, medium and low dose groups were significantly increased. Conclusion The ethyl acetate fraction of Arye red extract can improve the learning and memory abilities of AD rats induced by Aβ 25-35 and reduce the acetylcholinesterase activity in the brain.
