目的观察人参三七川芎醇提物延缓内皮细胞衰老的可能作用机制。方法实验分为衰老细胞组、自噬模型组、白藜芦醇组、人参三七川芎醇提物组,除衰老细胞组外,其余各组的衰老人心脏微血管内皮细胞(HCMEC)通过饥饿方法建立自噬模型。白藜芦醇组用10μmol/L白藜芦醇溶液培养,人参三七川芎醇提物组用100 mg/L人参三七川芎提取物溶液培养48 h。采用电子显微镜观察细胞中自噬小体的形态学改变,western blot法检测自噬蛋白Beclin1、LC3B表达情况。结果与自噬模型组比较,人参三七川芎醇提物组和白藜芦醇组细胞质中出现较多的独立双层及多层膜结构的自噬体。与自噬模型组比较,人参三七川芎醇提物组和白藜芦醇组Beclin1、LC3B蛋白表达明显增多(P<0.01),且白藜芦醇组Beclin1、LC3B蛋白的表达明显多于人参三七川芎醇提物组(P<0.01)。结论人参三七川芎醇提物可促进衰老内皮细胞自噬体的产生及自噬蛋白的表达,这可能是其延缓内皮衰老的重要机制之一。
Objective To observe the possible mechanism of ginseng extract of Chuanxiong Panax notoginseng which delays the senescence of endothelial cells. Methods The experiment was divided into senescent cell group, autophagy model group, resveratrol group, and ginseng extract of Chuanxiong notoginseng group. In addition to the senescent cell group, the senescent human cardiac microvascular endothelial cells (HCMEC) Establish an autophagy model. The resveratrol group was cultured with 10μmol / L resveratrol solution, the extract of Panax notoginseng chuanxiong ethanol group was treated with 100 mg / L ginseng extract of Panax notoginseng for 48 hours. The morphological changes of autophagosomes were observed by electron microscope. The expression of autophagy Beclin1 and LC3B were detected by western blot. Results Compared with the autophagy model group, there were more autophagosomes in the cytoplasm of the ginseng extract group and the resveratrol group. Compared with the autophagy model group, the expression of Beclin1 and LC3B protein in the extract of ginseng chuanxiong group and resveratrol group were significantly increased (P <0.01), and the expression of Beclin1 and LC3B protein in resveratrol group was significantly higher than that of ginseng Panax notoginseng ethanol extract group (P <0.01). Conclusion Ginseng extract of Chuanxiong notoginseng can promote the production of autophagosomes and the expression of autophagy in senescent endothelial cells, which may be one of the important mechanisms of its anti-aging activity.
