目的:优化复方扶芳藤胶囊的复合酶提取工艺。方法:采用单因素试验考察Celluclast~1.5L纤维素酶用量、Pectines XXL果胶酶用量、pH值、酶解温度、料液比和提取时间对人参皂苷Rb1转移率、总糖得率和干膏得率的影响,然后采用效应面法优化提取工艺。结果:优化的最佳工艺条件为:纤维素酶用量为1.0%、果胶酶用量为1.0%、pH值为5.2、酶解温度为56℃、料液比为1∶14.38、提取时间为4 h,人参皂苷Rb1的转移率为79.75%,总糖得率为13.57%,干膏得率为16.78%,实际OD值与理论值偏差为-2.51%。结论:纤维素酶联用果胶酶能高效提取复方扶芳藤胶囊有效成分,效应面法优化的提取工艺合理、可行。
Objective: To optimize the complex enzyme extraction process of Fufang Fufang capsule. Methods: Single factor experiments were conducted to investigate the effect of Celluclast ~ 1.5L cellulase dosage, Pectines XXL pectinase dosage, pH value, hydrolysis temperature, solid-liquid ratio and extraction time on the ginsenoside Rb1 transfer rate, The effect of paste yield, and then optimize the extraction process using the effect surface method. Results: The optimum conditions were as follows: the amount of cellulase was 1.0%, the amount of pectinase was 1.0%, the pH value was 5.2, the enzymolysis temperature was 56 ℃, the ratio of material to liquid was 1:14.38 and the extraction time was 4 h, ginsenoside Rb1 transfer rate of 79.75%, the total sugar yield was 13.57%, the dry paste yield was 16.78%, the actual OD value of the deviation from the theoretical value of -2.51%. CONCLUSION: Cellulase and pectinase can effectively extract the active ingredients of Fufang Futenng capsule, and the extraction process optimized by effect surface method is reasonable and feasible.
