目的应用新西兰大白兔建立慢性牙周炎(CP)合并动脉粥样硬化(AS)的动物模型,探讨CP对AS血管内皮功能的影响。方法 将32只雄性新西兰大白兔随机分为4组:对照组、CP组、AS组、CP+AS组。各组分别按照实验设计进行相应干预处理,至14周末处死动物,检测血清中血脂、内皮素1、一氧化氮含量,观察实验牙X线片及升主动脉的病理改变,测定主动脉壁E选择素表达的阳性面积百分比。结果 高脂喂养升高血清总胆固醇、三酰甘油、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C)水平(P<0.01),牙周炎升高血清三酰甘油水平(P<0.05)。对照组、CP组、AS组、CP+AS组血清内皮素1水平分别为(87.7±10.1)、(104.9±8.9)、(112.7±14.3)、(132.2±10.5)ng/L,血清一氧化氮水平分别为(31.83±4.21)、(28.34±4.29)、(27.75±2.94)、(25.59±3.03)μmol/L;析因设计资料方差分析表明,CP和AS均可升高血清内皮素1和升主动脉壁E选择素的表达水平,降低血清一氧化氮水平(均P<0.05),两种疾病合并未见明显协同效应(P>0.05)。结论 CP和AS均可导致血管内皮功能不全,两种疾病合并未见协同作用。
Objective To establish an animal model of chronic periodontitis (CP) combined with atherosclerosis (AS) in New Zealand white rabbits to investigate the effect of CP on the function of vascular endothelial cells. Methods Thirty-two male New Zealand white rabbits were randomly divided into 4 groups: control group, CP group, AS group and CP + AS group. The rats were sacrificed at the end of the 14th week. Serum lipids, endothelin-1 and nitric oxide were detected. The pathological changes of the experimental teeth and the ascending aorta were observed. Results High fat diet increased serum levels of total cholesterol, triglyceride, high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C) (P <0.05). The levels of endothelin 1 in the control, CP, AS and CP + AS groups were (87.7 ± 10.1), (104.9 ± 8.9), (112.7 ± 14.3) and (132.2 ± 10.5) ng / Nitrogen levels were (31.83 ± 4.21), (28.34 ± 4.29), (27.75 ± 2.94) and (25.59 ± 3.03) μmol / L, respectively. Variance analysis of factorial design data showed that both CP and AS increased serum endothelin 1 (P <0.05). There was no obvious synergistic effect between the two diseases (P> 0.05). Conclusions Both CP and AS lead to vascular endothelial dysfunction, and there is no synergistic effect between the two diseases.
