目的:评价芪玄益心胶囊对糖尿病(DM)大鼠的降糖作用,并探讨其作用机制。方法:以四氧嘧啶腹腔注射致胰腺损伤复制大鼠DM模型,将其随机分美吡达组、美吡达+芪玄益心胶囊高、中、低剂量组、模型组,并设空白对照组。观察各组模型血糖、糖化血清蛋白、胰腺白介素-1β(IL-1β)及其mRNA、核转录因子-κBp50(NF-κBp50)及其抑制蛋白(IκBа)水平,并与空白组对照。结果:模型组及各治疗组与空白组比较,血糖、糖化血清蛋白、IL-1β及其mRNA、NF-κBp50(细胞核)、IκBа(细胞浆)表达均显著增高(P<0.05)。美吡达+芪玄益心胶囊各剂量组与模型组比较,血糖、糖化血清蛋白、IL-1β及其mRNA、NF-κBp50、IκBа表达明显降低(P<0.05)。芪玄益心胶囊与美吡达比较,美吡达+芪玄益心胶囊高剂量组对胰腺IL-1β及其mRNA、NF-κBp50、IκBа改善优于美吡达(P<0.05)。结论:糖尿病大鼠胰腺中存在IL-1β、NF-κBp50的激活,芪玄益心胶囊对其有一定的抑制作用,推测抑制IL-1β-NF-κBp50(IκBа)这一途径的激活,是芪玄益心胶囊降低血糖,胰腺保护作用的重要机制之一。
Objective: To evaluate the hypoglycemic effect of Qixuanyixin Capsule on diabetic rats and to explore its mechanism of action. METHODS: Rat model of DM was induced by intraperitoneal injection of alloxan, and was randomly divided into pyestrazol group, mepyridam + guanxuanyixin capsule high, middle and low dose groups, model group, and blank control group. group. The levels of blood glucose, glycosylated serum protein, pancreatic interleukin-1β (IL-1β) and its mRNA, nuclear factor-κBp50 (NF-κBp50) and its inhibitory protein (IκBа) were observed in each group, and compared with the blank group. Results: The blood glucose, glycosylated serum protein, IL-1β and its mRNA, NF-κBp50 (nuclei) and IκBа (cytoplasm) expression were significantly higher in the model group and each treatment group compared with the blank group (P<0.05). Compared with the model group, the levels of blood glucose, glycosylated serum protein, IL-1β, mRNA, NF-κBp50, and IκBа were significantly decreased in each group (P<0.05). The comparison between Gengxuanyixin Capsule and Metopyol showed that the high-dose group of Metopyliden+Xuanxuanyixin Capsule had better improvement of pancreatic IL-1β and its mRNA, NF-κBp50, and IκBа (P<0.05). CONCLUSION: There is activation of IL-1β and NF-κBp50 in the pancreas of diabetic rats, and it is inhibited by Qixuanyixin Capsule. It is speculated that the activation of IL-1β-NF-κBp50 (IκBа) pathway is presumed to be Qi Xuan Yi Xin Capsule reduces blood sugar, one of the important mechanisms of pancreatic protection.
