目的观察小儿平喘方对哮喘模型大鼠血清中γ-干扰素(IFN-γ)、白细胞介素-4(IL-4)含量、IFN-γ/IL-4比值的影响,从而探讨小儿平喘方治疗支气管哮喘的药效机制。方法选取SD大鼠32只,随机分为4组:正常对照组、哮喘模型组、小儿平喘方小剂量组(小剂量组)和小儿平喘方大剂量组(大剂量组),每组8只,采用卵清蛋白(OVA)致敏和雾化吸入激发制备大鼠哮喘模型,用双位点夹心酶联免疫吸附试验测定各组大鼠血清中IL-4和IFN-γ含量。结果与正常对照组比较,哮喘模型组大鼠血清IFN-γ的含量与IFN-γ/IL-4比值均降低(P<0.05),而IL-4的含量则显著升高(P<0.05);与哮喘模型组比较,大、小剂量组大鼠血清IFN-γ的含量与IFN-γ/IL-4比值均显著升高(P<0.01),而IL-4的含量则降低(P<0.05)。结论小儿平喘方能显著降低支气管哮喘模型大鼠血清IL-4的含量,提高IFN-γ的含量及失衡的IFN-γ/IL-4比值,从而调节免疫功能,减轻炎症反应,控制哮喘的发作。
Objective To observe the effect of Xiaoer Pingchuan Recipe on the serum levels of IFN-γ, IL-4 and IFN-γ / IL-4 in asthmatic rats. Asthma recipe for the treatment of bronchial asthma efficacy mechanism. Methods Thirty-two SD rats were randomly divided into 4 groups: normal control group, asthma model group, low dose Pingchuan Pingchuan recipe group (small dose group) and high dose Pingchuan recipe group (high dose group) Eight rats were sensitized with ovalbumin (OVA) and challenged by inhalation of atomization to prepare the model of asthma in rats. The levels of IL-4 and IFN-γ in sera of rats in each group were measured by double-site sandwich enzyme-linked immunosorbent assay. Results Compared with the normal control group, the serum levels of IFN-γ and IFN-γ / IL-4 in asthmatic model group were significantly decreased (P <0.05), while the levels of IL-4 were significantly increased (P <0.05) Compared with the asthma model group, the levels of IFN-γ and IFN-γ / IL-4 in the large and small dose groups were significantly increased (P <0.01), while the levels of IL-4 were decreased 0.05). Conclusion Pediatric asthma can significantly reduce the level of serum IL-4, increase the level of IFN-γ and the imbalanced IFN-γ / IL-4 ratio in asthmatic rats, so as to regulate immune function, reduce inflammation and control asthma attack.
