目的观察双氢青蒿素(DHA)对卵巢癌细胞株H0-8910裸鼠皮下移植瘤的作用及p-Akt表达,探讨其机制。方法将H0-8910细胞注入裸鼠,建立裸鼠移植瘤模型,观察干预后各组移植瘤抑瘤率,免疫组化检测p-Akt的活性变化。结果①病理学观察见细胞有局灶性坏死,细胞变大,胞质疏松,核深染,可见核固缩、核溶解、核碎裂,染色质成团状。②DHA对裸鼠移植瘤生长具有明显的抑制作用,并具有浓度依赖性。③免疫组化检测p-Akt活性发现随着双氢青蒿素剂量的增加p-Akt表达减弱。结论双氢青蒿素对人卵巢癌细胞株HO-8910裸鼠皮下移植瘤有明显的抑制作用,其机制与下调p-Akt的表达可能有关。
Objective To observe the effect of dihydroartemisinin (DHA) on the subcutaneous xenografts of ovarian cancer cell line H0-8910 in nude mice and its mechanism. Methods H0-8910 cells were injected into nude mice to establish a nude mouse xenograft model. The inhibitory rate of tumor in each group was observed and the activity of p-Akt was detected by immunohistochemistry. Results ① Pathological observation showed that the cells had focal necrosis, enlarged cells, loose cytoplasm, deep stained nuclei, nuclear pyknosis, nuclear lysis, nuclear fragmentation and chromatin mass formation. ② DHA has a significant inhibitory effect on the growth of xenografts in nude mice, and has a concentration-dependent manner. ③ Immunohistochemical detection of p-Akt activity found that with the increase of dihydroartemisinin, the expression of p-Akt was weakened. Conclusions Dihydroartemisinin can significantly inhibit the subcutaneous xenografts of human ovarian cancer cell line HO-8910 in nude mice, and its mechanism may be related to the down-regulation of p-Akt expression.
