目的 应用重组痘苗病毒介导的IL-2对小鼠Lewis肺癌直接进行局部转染,并对其疗效进行观察和评价。方法 本项研究采用缺陷型重组痘苗病毒为载体,经同源重组,筛选出IL-2基因的rVV-IL-2,体外转染小鼠Lewis肺癌,对其疗效进行观察。结果 短期内即可检出IL-2的表达,并且免疫原性也得到提高。用该载体以in vivo方式原位转染荷瘤小鼠局部;激活了小鼠体内的CTL、LAK及NK细胞,显著抑制了小鼠肿瘤的生长,部分小鼠未能形成肿瘤。结论 提示重组痘苗病毒为载体介导的IL-2基因治疗具有良好的前景。
OBJECTIVE: To directly transfect mouse Lewis lung carcinoma with recombinant vaccinia virus IL-2 and observe its therapeutic effect. Methods The recombinant vaccinia virus was used as a vector to screen rVV-IL-2 gene of IL-2 gene by homologous recombination. The transfection of Lewis lung carcinoma in vitro was performed and the therapeutic effect was observed. As a result, the expression of IL-2 can be detected in a short period of time, and the immunogenicity is also improved. In vivo, the vector was used to transfect the tumor-bearing mice in situ. Activation of CTL, LAK and NK cells in mice significantly inhibited tumor growth in mice and some mice failed to form tumors. Conclusions Recombinant vaccinia virus vector-mediated IL-2 gene therapy has a good prospect.
