①目的探讨青紫薯色素对自然衰老大鼠体内抗氧化能力的影响。②方法以健康青年Wistar大鼠为正常对照组,50只自然衰老Wistar大鼠分为模型组,青紫薯色素高、中、低剂量(1 500、150、15 mg/kg)组和维生素E(2 mg/kg)组。正常对照组和模型组给予生理盐水20 mL/kg,其他各组每日灌胃1次,每次3 mL,连续给药30 d。应用生化方法检测青紫薯色素对自然衰老Wistar大鼠体内两种脏器(脑、心脏)组织匀浆的超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-px)和总抗氧化能力(T-AOC)的影响,并探讨青紫薯色素剂量与三种抗氧化指标之间的相关关系。③结果自然衰老大鼠脑、心脏组织匀浆中SOD、GSH-px和T-AOC活性均显著降低,青紫薯色素可以提高衰老大鼠组织匀浆SOD、GSH-px的活性,增强衰老大鼠机体的T-AOC,与模型组比较,差异具有显著统计学意义(F=9.05~44.09,q=2.86~12.10,P<0.05)。3种抗氧化指标中,脑组织中SOD、GSH-px和T-AOC水平随青紫薯色素剂量的增加而增高(r=0.87~0.93,P<0.05);而心脏组织中SOD、GSH-px和T-AOC水平与青紫薯色素的剂量不呈正相关关系(r=0.43~0.51,P>0.05)。④结论青紫薯色素可以通过有效调节自然衰老大鼠体内的氧化水平,延缓衰老进程,剂量依赖性地提高脑组织的抗氧化能力。
①Objective To investigate the effect of purple sweet potato pigment on the antioxidant capacity in the natural aging rats. Methods The healthy Wistar rats were used as normal control group. Fifty naturally aged Wistar rats were divided into model group, high, medium and low doses of purple sweet potato pigment (1 500, 150, 15 mg / kg) and vitamin E 2 mg / kg) group. Normal control group and model group were given normal saline 20 mL / kg, the other groups were given gavage once a day, 3 mL each for 30 days. Biochemical methods were used to detect the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-px), and glutathione peroxidase (GSH-px) in the tissue homogenates of two organs (brain and heart) Total antioxidant capacity (T-AOC), and explore the relationship between the dose of purple sweet potato pigment and three antioxidant indicators. ③ Results The activities of SOD, GSH-px and T-AOC in brain tissue and heart homogenate of rats with natural aging were significantly decreased. Potato tuber pigment could increase the activity of SOD and GSH-px in the tissue of aging rats, The difference of T-AOC between the model group and the model group was statistically significant (F = 9.05 ~ 44.09, q = 2.86 ~ 12.10, P <0.05). The levels of SOD, GSH-px and T-AOC in brain tissue increased with the increase of purple pigment dose (r = 0.87-0.93, P <0.05), while SOD, GSH-px There was no positive correlation between T-AOC level and the dose of purple pigment (r = 0.43-0.51, P> 0.05). ④ Conclusion Potato purple pigment can effectively slow the aging process and increase the antioxidant capacity of brain tissue in a dose-dependent manner by effectively regulating the oxidation of natural aging rats.
