以‘太平红’牡丹为材料,研究了不同灭菌方式、抗褐化物质、激素配比、培养基、生根培养条件对牡丹组织培养的影响。结果表明:以0.1%升汞液消毒8 min,再用7%次氯酸钠溶液消毒8 min灭菌效果最佳,鳞芽、土芽和种子的污染率分别为0、4.5%和22.0%;在改良MS培养基中联合添加PVPP 0.5 g/L和AC 0.5 g/L可有效减少牡丹外植体的褐化;改良MS+BA 1.0 mg/L+NAA 0.2 mg/L+GA 0.3 mg/L+CH 0.5 g/L+PVPP0.5 g/L+AC 0.5 g/L适宜牡丹鳞芽的诱导;MS+BA 1.5 mg/L+NAA 0.2 mg/L+PVPP 1.0g/L适宜于牡丹芽的增殖;1/2MS+IBA 5 mg/L+AC 0.5 g/L适用于牡丹试管苗生根。
The effects of different sterilization methods, anti-browning substances, hormone ratio, medium and rooting culture conditions on the tissue culture of Peony were studied using ’Taipinghong’ Peony as material. The results showed that the disinfection with 0.1% mercuric chloride solution for 8 min and the disinfection with 7% sodium hypochlorite solution for 8 min resulted in the best sterilization effect. The contamination rates of scale bud, soil bud and seed were 0, 4.5% and 22.0% respectively. MS supplemented with 0.5 g / L PVPP and 0.5 g / L AC could effectively reduce the browning of peony explants; MS + BA 1.0 mg / L + NAA 0.2 mg / L + GA 0.3 mg / L + 0.5 g / L + PVPP0.5 g / L + AC 0.5 g / L was suitable for peony bud induction; MS + BA 1.5 mg / L + NAA 0.2 mg / L + PVPP 1.0 g / 1 / 2MS + IBA 5 mg / L + AC 0.5 g / L Suitable for rooting of peony plantlets.
