目的探讨神经生长因子(nerve growth factor,NGF)及其酪氨酸激酶受体A(TrkA)与人胆管癌神经浸润、淋巴转移和病理类型的关系。方法采用免疫组织化学技术,检测92例人胆管癌组织中β-NGF和TrkA的表达情况,Spearman’s等级相关检验β-NGF的表达与胆管癌神经浸润、淋巴转移以及病理分型的关系。结果β-NGF和TrkA均在胆管癌组织中高表达,二者存在相关性,且在神经浸润组和无神经浸润组的表达差异有统计学意义;NGF阳性表达随着肿瘤恶性程度的增高而增高,且与肿瘤的淋巴转移密切相关。TrkA还表达于神经鞘膜。结论NGF与胆管癌神经浸润密切相关。NGF可能通过旁分泌途径与TrkA作用促使癌细胞沿神经转移;通过自分泌作用促进胆管癌细胞增殖。
Objective To investigate the relationship between nerve growth factor (NGF) and its tyrosine kinase receptor A (TrkA) and neural invasion, lymphatic metastasis and pathological types of human cholangiocarcinoma. Methods Immunohistochemical technique was used to detect the expression of β-NGF and TrkA in 92 cases of human cholangiocarcinoma. Spearman’s rank correlation test was used to examine the relationship between β-NGF expression and nerve invasion, lymphatic metastasis, and pathological type of cholangiocarcinoma. Results The expressions of β-NGF and TrkA were highly expressed in cholangiocarcinoma and they were correlated with each other. The expression of NGF and TrkA were significantly different in the neural infiltration group and the non-neural infiltration group. The positive expression of NGF increased with the increase of the malignant degree of the tumor. , and is closely related to the lymphatic metastasis of the tumor. TrkA is also expressed in the nerve sheath. Conclusion NGF is closely related to nerve invasion of cholangiocarcinoma. NGF may promote the metastasis of cancer cells along the nerves through the paracrine pathway and TrkA; autocrine promotes proliferation of cholangiocarcinoma cells.
