目的:应用蛋白芯片技术筛选并分析参苈加颗粒对心力衰竭大鼠心肌单核细胞趋化蛋白-1(MCP-1)表达的影响。方法:将70只清洁级Wistar大鼠随机抽取空白组15只,其余用阿霉素造模,将成模大鼠随机分为模型组和治疗组,分别给予参苈加颗粒和0.9%氯化钠溶液灌胃,每日1次。4周后取大鼠左心室组织。结果:空白组心肌正常;模型组心肌纤维增粗、紊乱,间质增生,少量炎症细胞浸润;治疗组心肌排列较整齐,间质无明显增生,未见炎症细胞浸润。蛋白芯片显示,与空白组相比,模型组的表达下调;与模型组比较,治疗组的表达上调;治疗组与空白组比较无表达差异。结论:参苈加颗粒能够通过上调MCP-1的表达,对心力衰竭大鼠的心肌起到保护作用。
OBJECTIVE: To screen and analyze the effect of Shenjuan Granule on the expression of MCP-1 in myocardium of rats with heart failure by protein chip technology. Methods: Seventy clean Wistar rats were randomly divided into blank group (n = 15) and adriamycin-supplemented group (n = 10). The model rats were randomly divided into model group and treatment group. Solution gavage, 1 day. After 4 weeks, rats’ left ventricular tissue was taken. Results: The myocardium in the blank group was normal. The myocardial fibers in the model group were thicker and disordered, interstitial hyperplasia and inflammatory cell infiltration. In the treatment group, the myocardial tissues were arranged neatly with no interstitial hyperplasia and no inflammatory cell infiltration. Protein chip showed that compared with the blank group, the expression of the model group was down-regulated; compared with the model group, the expression of the treatment group was up-regulated; there was no difference between the treatment group and the blank group. Conclusion: Shenwei granule can protect myocardium of rats with heart failure by increasing the expression of MCP-1.
