[目的]探讨芪丹煎对家兔动脉粥样硬化干预作用的分子机制。[方法]建立家兔动脉粥样硬化模型,比较对照组、高脂组、西药组和芪丹煎组动脉粥样硬化斑块形成的变化;检测各组血清血脂水平;免疫组化结合RT-PCR法分析Bcl-2、Bax在粥样硬化斑块中的表达。[结果]芪丹煎组血脂水平和动脉粥样斑块面积低于高脂组(P﹤0.01);免疫组化显示,高脂组Bcl-2、Bax表达与正常组差异有统计学意义。而芪丹煎的干预可以调节Bax/Bcl-2阳性细胞表达率。[结论]芪丹煎可抑制动脉粥样硬化斑块的形成;调节Bcl-2/Bax表达可能是抗动脉粥样硬化的机制之一。
[Objective] To investigate the molecular mechanism of Qidan Jian intervention on atherosclerosis in rabbits. [Method] The model of atherosclerosis in rabbits was established. The changes of atherosclerotic plaque formation in control group, high-fat group, western medicine group and Qidanjian group were compared. Serum lipid levels were measured in each group. Immunohistochemistry and RT- PCR analysis of Bcl-2, Bax expression in atherosclerotic plaque. [Results] The serum lipid and atherosclerotic plaque area in Qidanjian decoction group were lower than those in the high-fat group (P <0.01). Immunohistochemistry showed that the expression of Bcl-2 and Bax in hyperlipidemia group was significantly different from that in the normal group. The QD decoction can regulate the expression of Bax / Bcl-2 positive cells. [Conclusion] QD can inhibit the formation of atherosclerotic plaque. Regulation of Bcl-2 / Bax expression may be one of the mechanisms of anti-atherosclerosis.
