油体是油料作物种子储存脂肪的细胞器,油体膜蛋白是构成油体的重要组分,对维持油体大小和稳定起着重要作用。本研究以油体膜蛋白为对象,克隆了凤丹的油体膜蛋白基因(OLE),其cDNA全长783 bp,开放阅读框长507 bp,共编码169个氨基酸,蛋白的分子量为17.48 k D,故将该基因命名为PoOLE17.5。生物信息学分析显示,该基因编码的蛋白包含2个亲水性区域和1个疏水性区域,并具有1个保守结构域“Oleosin domain”。而后的表达模式显示,PoOLE17.5在凤丹种子发育过程中的表达水平逐渐上升,在种子接近成熟时表达水平最高,而后略有下降。这一结果有利于今后通过基因工程手段调控凤丹油体发育。
Oil body is an organelle for storing fat in oilseed crops. Oil membrane protein is an important component of oil body and plays an important role in maintaining oil body size and stability. In this study, the oil membrane protein of P. oleifera cloned the oil body membrane protein gene (OLE), its full-length cDNA was 783 bp, the open reading frame length of 507 bp, encoding a total of 169 amino acids, the molecular weight of the protein was 17.48 k D, so the gene named PoOLE17.5. Bioinformatics analysis showed that the protein encoded by this gene contains two hydrophilic regions and one hydrophobic region, and has a conserved domain “Oleosin domain ”. The expression pattern of PoOLE17.5 showed that the expression level of PoOLE17.5 in the process of seed development of Fengdan was gradually increased, and the expression level of PoOLE17.5 was the highest when the seed was near maturity, then decreased slightly. This result is conducive to the future through genetic engineering means to regulate the development of Fengyan oil body.
