目的本文主要研究近几年来NCBI数据库最新释放出来的8种新型HPV即HPV(126,135,136,137,140,144,154,166),对其进行分子进化树分析,进行分类并设计特异型引物。方法 NCBI检索8种新型HPV的基因组序列,然后用Blast相似性比较得到与其L1高度相似的HPV,其次选择HPVαβγ各属具代表性的HPV,最后用Clustal W对以上所有58种HPV L1区氨基酸序列进行比较分析,构建分子进化树;进一步用Primer-Blast对8种新型HPV进行特异型引物设计。结果系统进化树分析得出8种HPV均属于γ属,利用Primer-Blast分析得到这8种HPV特异型引物。结论 HPV126、HPV135、HPV136、HPV137、HPV140、HPV144、HPV154、HPV166均为γ属。
OBJECTIVE In this paper, the eight new types of HPV (126,135,136,137,140,144,154,166) newly released from NCBI database were studied in recent years. They were analyzed by molecular phylogenetic tree, classified and designed specific primers. Methods Genomic DNA sequences of eight novel HPV types were searched by NCBI. HPVs with high similarity to L1 were obtained by Blast similarity. Second, HPVs were selected from each genus of HPVαβγ. Finally, Clustal W was used to analyze the amino acid sequence of all 58 HPV L1 regions The molecular phylogenetic tree was constructed and compared with Primer-Blast. Eight novel HPV-specific primers were designed. Results Phylogenetic tree analysis showed that all eight HPVs belonged to γ genus, and the eight HPV-specific primers were obtained by Primer-Blast analysis. Conclusion HPV126, HPV135, HPV136, HPV137, HPV140, HPV144, HPV154 and HPV166 are all γ genera.
