本文报道了小鼠U_(14)肿瘤组织环—磷酸腺苷磷酸二酯酶(PDE)活力的变化。PDE活力测定系采用~3H-cAMP做底物,用PDE和蛇毒两步水解法,用液闪仪测定~3H-腺苷生成量,计算出PDE活力。实验结果表明,小鼠U_(14)肿瘤组织与对照组比较,PDE活力明显升高,P<0.01。肿瘤组织PDE活力升高是大多数恶性肿瘤的瘤细胞环—磷酸腺苷(cAMP)含量低下的主要原因,而cAMP含量降低又与肿瘤的发生有密切关系,所以深入研究PDE变化及其本质,可能为肿瘤发病机理提供线索。应用PDE抑制剂,提高肿瘤组织cAMP含量,亦可能有助于肿瘤治疗,因此有可能把PDE做为寻找新药的一个特异性靶子,用来做为筛选肿瘤治疗药物的生化指标之一。
In this paper, we report the changes in the activity of cyclic adenosine phosphodiesterase (PDE) in mouse U_(14) tumor tissue. PDE activity was measured using ~3H-cAMP as a substrate, and PDE and snake venom two-step hydrolysis were used. The amount of ~3H-adenosine was measured using a liquid scintillation counter to calculate the PDE activity. The experimental results showed that compared with the control group, the PDE activity of the mouse U_(14) tumor tissue was significantly increased, P<0.01. The increase in PDE activity in tumor tissue is the main reason for the low level of cyclic AMP (cAMP) in tumor cells of most malignant tumors. The decrease in cAMP content is closely related to the occurrence of tumors. Therefore, the changes in PDE and its nature are deeply studied. It may provide clues to the pathogenesis of the tumor. The use of PDE inhibitors to increase the cAMP content of tumor tissue may also contribute to tumor treatment. Therefore, it is possible to use PDE as a specific target for finding new drugs to be used as one of the biochemical indicators for screening tumor treatment drugs.
