目的探讨葛根主要成分葛根总黄酮对急性早幼粒细胞白血病(APL)细胞株NB4增殖及凋亡的影响。方法采用MTT比色法检测葛根总黄酮对NB4细胞增殖抑制率;光学显微镜及荧光显微镜观察细胞形态改变;AnnexinV/PI双染法检测细胞凋亡率;DNAPI染色法分析细胞周期及亚二倍体峰。结果12.5~200μg/ml葛根总黄酮均能抑制NB4细胞的增殖。光镜及荧光显微镜下可见核固缩、凋亡小体等典型的细胞凋亡改变;AnnexinV+/PI-细胞呈时间-剂量依赖性增加;DNAPI染色法发现细胞亚二倍体比例增加,G1期比例下降、S期比例增高。结论葛根总黄酮能有效抑制NB4细胞增殖、阻滞细胞周期进程、诱导细胞凋亡。
Objective To investigate the effects of Pueraria lobata (Pueraria lobata) root extract on the proliferation and apoptosis of acute promyelocytic leukemia (APL) cell line NB4. Methods MTT colorimetry was used to detect the inhibitory rate of total flavonoids of Pueraria lobata on the proliferation of NB4 cells. The morphological changes of the cells were observed by light microscopy and fluorescence microscope. The apoptosis rate of the cells was detected by Annexin V / PI double staining. peak. Results Total flavonoids of Pueraria lobata at concentrations of 12.5 ~ 200μg / ml could inhibit the proliferation of NB4 cells. Apoptosis of apoptotic cells was observed under light microscope and fluorescence microscope. AnnexinV + / PI-cells increased in a time-and dose-dependent manner. The percentage of sub-diploid cells was increased by DNAPI staining, Proportion decreased, S-phase ratio increased. Conclusion Pueraria total flavonoids can effectively inhibit NB4 cell proliferation, arrest cell cycle progression and induce apoptosis.
