目的观察不同浓度可吸入颗粒物(PM2.5)对T2DM大鼠凝血纤溶因子的影响。方法采用10%水合氯醛腹腔麻醉,气管内滴注法染毒,24h后处死大鼠,股动脉收集血液,检测凝血酶原时间(PT)、部分凝血活酶时间(APTT)、纤维蛋白原(FIB)、纤溶酶原激活物抑制剂-1(PAI-1)水平。结果不同剂量染毒组PT、PAI-1、FIB与其生理盐水组比较,差异有统计学意义(P<0.05)。滴注相同剂量PM2.5的T2DM大鼠PT、PAI-1、FIB较Wistar大鼠高(P<0.05)。结论不同浓度PM2.5会导致Wistar大鼠凝血纤溶因子的改变,PM2.5对T2DM大鼠凝血纤溶系统的影响更加明显,可能加速糖尿病的进程。
Objective To observe the effects of different concentrations of inhalable particles (PM2.5) on coagulation and fibrinolytic factors in T2DM rats. Methods The rats were anesthetized by intraperitoneal instillation of 10% chloral hydrate and endotracheally instilled. After 24h, the rats were sacrificed and the blood was collected from the femoral artery. Prothrombin time (PT), partial thromboplastin time (APTT), fibrinogen (FIB), plasminogen activator inhibitor-1 (PAI-1) levels. Results Compared with saline group, PT, PAI-1 and FIB in different dose groups showed significant difference (P <0.05). The PT, PAI-1 and FIB in T2DM rats with the same dose of PM2.5 were significantly higher than those in Wistar rats (P <0.05). Conclusion Different concentration of PM2.5 can cause changes of coagulation fibrinolytic factor in Wistar rats. PM2.5 has a more obvious effect on coagulation and fibrinolytic system of T2DM rats, which may accelerate the process of diabetes.
